CarcinogenicityAcceptedBeyond the letter
No dedicated carcinogenicity studies with aprocitentan; mouse and rat studies of parent drug macitentan used instead
Why: Aprocitentan is a major metabolite of macitentan, at least 38% of total exposure in those studies · Pharmacology or kinetics · Weight of evidence
“No dedicated carcinogenicity studies were conducted with aprocitentan. Aprocitentan is a major metabolite of macitentan, and its exposure represented ≥38% of the total exposure to macitentan and aprocitentan in the macitentan carcinogenicity studies. Therefore, the macitentan carcinogenicity studies, conducted in B6C3F1 mice and Wistar rats, were considered acceptable to assess the carcinogenic potential of aprocitentan.”
